Following the death of her husband, she used her inheritance to establish a commercial empire. After fleeing the horrors of the Inquisition in Portugal, she began practicing Judaism openly and expanded her business interests to Brussels, Venice and the Ottoman Empire. Dona Gracia supported Jews throughout Europe and protected conversos. In response to the execution of conversos in Ancona, she organized a trade embargo on its port, bringing commercial life in the city to a standstill.
